EMSA showed that BldD binds specifically to the whiG promoter region. DNase I footprinting identified that BldD protected two sites in the whiG promoter. S1 nuclease assay experiment compared the expression levels of whiG in the wild type and bldD mutant strains and the results suggested that BldD repressed whiG expression. Northern blot analysis confirmed BldD-mediated regulation. Similarly, BldD-mediated regulation of bldN and bdtA was shown.
