LacZ fusion assays determined that FleQ is an activator of fliE, flgG, flgB, flhF, fliL, and fliQ expression. Multiple sequence alignment of the upstream regions of genes of interest yielded a consensus sequence. Consensus search focusing on these six FleQ dependent promoters yielded a weak consensus binding sequence upstream of each RpoN binding site. Insertional mutagenesis showed that deletion of the predicted FleQ binding site from the fliE promoter abolished the promoter activity
