Experimental Process

2D gel electrophoresis was used to compare the protein patterns of the P. aeruginosa narL mutant with that of the wild-type PAO1. Northern blot analysis in wild type and narL mutant strains showed that NarL represses arcDABC expression. LacZ reporter assays in the wild type and narL mutant confirmed arcDABC repression by NarL. Site-directed mutagenesis of the putative NarL binding site in conjunction with arcD-lacZ fusion assays confirmed direct repression of arcDABC by NarL.
