Crp-mediated activation of phhAB operon was confirmed by phhAB-lacZ fusion assays in the wild-type and Crp and Cya mutant strains. EMSA revealed that the Crp bound to the phhAB promoter when cAMP was present. DNase I footprinting identified the Crp binding region in the phhAB promoter. Random mutagenesis of the putative Crp binding site confirmed that Crp binding to its target site is specific.
