Experimental Process

Site-directed mutagenesis combined with lacZ transcriptional fusion narrowed down the region of cop promoter necessary for copper inducible expression. EMSA determined that CopR binds specifically to the cop promoter. DNase I footprinting localized the CopR binding site.
