EMSA confirmed that CmeR specifically binds to the promoter region of cmeABC. Specific CmeR binding was verified by performing a gel mobility shift assay with a series of PCR products whose sequences span different portions of the region between cmeR and cmeABC. Site-directed mutagenesis in conjunction with EMSA and lacZ reporter fusion assay showed that the single deletional mutation increased the level of transcription of cmeABC.
