Computer analysis of the ptxR-promoter region revealed a potential Vfr binding site. EMSAs showed that Vfr binds to this region. Vfr binding site was then localized by performing DNase I footprinting. The protected site contained the putative Vfr binding site. Then Vfr was shown to be an activator of ptxR expression by comparing the results of the lacZ fusion assays in WT and vfr deletion mutant.
