Experimental Process

Fur-mediated regulation was shown by performing complementation assays. Site-directed mutagenesis of the putative Fur box in conjunction with nrdHIEF-lacZ reporter fusion confirmed Fur-mediated regulation of nrdHIEF. Specific binding of Fur to its target promoter was confirmed by performing EMSAs with a wild-type promoter and a promoter containing an altered Fur box.
