Experimental Process

Site directed mutagenesis of the C-terminal end of TraR abolished transcription of traI and traM. Results were supported by lacZ analyses. EMSA confirmed that TraR binds to tra box II and tra box IV. Western blot analyses quantified TraR accumulations of the various mutants in vivo.
