Experimental Process

The DNA-binding domain of MogR and its binding site in the flaA promoter were identified with EMSA on multiple fragment sizes and DNAse footprinting on the flaA promoter. Western blots were used to validate repression of flaA by the MogR binding domain. The MogR N-terminal region containing the DNA binding domain was co-crystallized with the flaA promoter region, showing specific contacts of the dimer with the DNA site and recognition of bend in the central region.
