Experimental Process

A consensus search found potential binding sites of CiaR in the S.mutans genome. DNase I footprinting determined a binding site at the ciaX gene. Site directed mutagenesis with EMSA confirmed the functionality proposed CiaR binding site. 5' RACE determined the transcription start site of the ciaX gene. DNA-array expression analysis determined that CiaR represses the ciaX gene.
