Experimental Process

DNA microarray analyses showed that the expression of 359 genes was different in the bldD mutant compared to the wild type strain. ChIP-chip experiments identified ca. 172 peaks throughout the S. coelicolor genome. MEME motif discovery tool identified a 15 bp BldD consensus sequence. DNase I footprinting carried out on the pepA-pepA2 intergenic region, cdgA, bldA and fitZ promoters, showed that in each case BldD protected a region containing a BldD box. EMSA confirmed the that BldD bound to the identified BldD box in the bldA promoter. The publication provided only the BldD binding sites upstream of genes involved in sporulation and/or antibiotic production.
