Experimental Process

qPCR analysis determined that the expression of the gal operon in the ccpA mutant was upregulated 5-fold compared to the wild type strain when cells grew in a mixture of glucose and galactose. Similarly, manL-luc reporter fusion determined that manL expression was 2-fold higher in the ccpA mutant than in the wild type strain. Visual inspection of the promoter regions of these genes identified CcpA binding motifs. EMSA confirmed that CcpA binds specifically to the manL and gal promoters.
