Experimental Process

CatR regulation of the pheBA operon in Pseudomonas putida was investigated as it related to an internal binding site (IBS), identified by DNase I protection in an earlier publication. Deletion analysis and site-directed mutagenesis demonstrated reduced pheBA expression so long as both the recognition binding site (RBS) and activation binding site (ABS) were intact.
