Experimental Process

DNA microarray analysis identified ArgR regulated genes in Pseudomonas aeruginosa PAO1. LacZ fusion assays using an argR knockout mutant were performed for all genes whose regulation by ArgR was previously unconfirmed, identifying ArgR activation of PA1818, PA1971, PA2024, PA3934, and the PA5152-5155 operon, and ArgR repression of argG, gdhA and the gltBD operon. EMSA confirmed the binding of ArgR to the promoter regions of argG, PA1971, PA2042, PA3934 and PA5152. Multiple sequence alignment of the promoters of the genes of interest and previously identified ArgR regulated promoters localized the ArgR binding sites and deduced an ArgR binding consensus sequence.
