Experimental Process

GFP promoter fusion assay, confirmed by beta-galactosidase reporter assay, indicated regulation of promoters in response to hypoxia. Transcription start points determined by primer extension. Site-directed mutagenesis inactivated suspected binding sites and expression assays confirmed reduced expression in response to hypoxia. DevR binding sites confirmed via DNAse I footprinting; 3 sites found to bind.
