Experimental Process

Putative Fur-binding sites in the genome of C. crescentus were detected by using a position weight matrix approach. EMSA validated that Fur binds to its target promoter region. LacZ reporter assays validated Fur-mediated expression regulation. Site-directed mutagenesis of Fur-binding sites confirmed specific binding. DNase I footprinting was used to locate Fur binding sequence.

Gene Regulation

Regulated genes for each binding site are displayed below. Gene regulation diagrams show binding sites, positively-regulated genes, negatively-regulated genes, both positively and negatively regulated genes, genes with unspecified type of regulation. For each indvidual site, experimental techniques used to determine the site are also given.
