Experimental Process

The TSS was identified by primer extension. VpsT sites in the VpsL promoter were identified by DNAse footprinting and their specificity validated by EMSA with mutated sites and Lux reporter assays with same site mutants and promoters truncated at several distances from the TSS. The motif was inferred by motif discovery on biofilm formation-related promoters and its stability validated by comparative genomics on V. cholerae isolates.
