Experimental Process

sdiA expression was shown to be reduced by 30% in leuO deletion mutant compared to the wild-type strain. EMSA confirmed that LeuO regulated sdiA expression directly. Footprinting further confirmed the EMSA results by showing that LeuO bound to two sites which overlap with the -35 and -10 sequences in the sdiA promoter.
