Experimental Process

A spx:bgaB fusion assay demonstrated repression of spx by YodB. Primer extension and In vitro transcription assays validated this claim. Site directed mutagenesis of the upstream region of spx resulted in derepression. DNase I footprinting localized the binding site. Site directed mutagenesis of this identified site confirmed its role in repression.
