LeuO-regulated proteins were identified by MALDI-TOF Mass Spectrometry. CAT reporter assays showed that LeuO activated the STY3070-STY3064 operon transcription and repressed the expression of tpx, ompX, and STY1978. Primer extension analysis was used to determine the transcription start sites of LeuO-regulated genes. DNase I footprinting determined the binding regions involved in LeuO-dependent regulation of STY3070, assT, ompS1, ompS2, tpx, ompX, and STY1978. The authors employed pattern discovery algorithms to identify LeuO consensus binding site and reported that LeuO binding sites are not sequence specific and may not be easily represented with a consensus.
