Experimental Process

DNase I footprinting showed that phosphorylated CtrA binds to its target sites with higher affinity than unphosphorylated CtrA. Mutagenesis of the CtrA binding sites in conjunction with DNase I footprinting showed that the whole site is required for phosphorylation-dependent binding.
