torECAD transcription start site was identified by primer extension analysis using RNA prepared from cells grown anaerobically with TMAO. torEC-lacZ fusion assays verified TorR-dependent regulation of torECAD. EMSA confirmed that this regulation is direct. DNase I footprinting localized the TorR binding site in the torECAD promoter. Deletion of the TorR box resulted in reduced expression and loss of binding.
